Also should talents like mixing/mastering be placed near the end when all stems or seps are in. Lastly, should stems be marked production ready by the Project Manager once they are in?

Should a listed talent in a project be removed once filled? Definitely yes on this one. We expect project managers to maintain their project, and that especially includes updating of talent requirements of the project for the reasons you have mentioned. I believe this is briefly mentioned in the Project Settings area currently. Once a member has auditioned for the part, and has been accepted into the project, the talent should be removed as soon as possible.
Should talents like mixing/mastering be placed near the end when all stems or seps are in? This is generally a good idea; though sometimes project managers might want to recruit for the position early on. Bearing in mind, however, that nothing can be either mixed or mastered until all seps are in! So posting for these particular talents would ideally be done at the time its needed, at the end as you suggest.
Should stems be marked production ready by the Project Manager once they are in? Tracks should be marked as 'production ready' only when they are completed. Once marked, the mixer knows that they are good to go and that there won't be any changes. Once tracks are marked as production ready, the collaborator won't be able to upload a new version of the track/make changes.
